A simple, fast, high-throughput, modified method for the simultaneous determination of citrate and oxalate in a single purified urine sample using two commercially available kits

Background: Urine citrate and oxalate levels are risk markers for renal stone formation. measurements prone to interferences linearity issues. This study investigates modification of sample preparation citric acid assay using the pH adjusted purified urine used analysis. Methods: samples were diluted 1:1 with diluent (Trinity Biotech Oxalate), vortexed between 6 8. Diluted transferred into purifier tubes vortexed, placed on a roller 5min, spun, filtered, followed by citrate(Enzytec Liquid citrate) oxalate(Trinity Oxalate) analysis 12min run time per 20 (Indiko- Plus platform). Results: Within-run imprecision this modified method was 3.94% CV (0.47 mmol/L) 1.02% (2.09 mmol/L). For 2.98% (0.28 0.86% (1.00 Linearity confirmed up 4 mmol/L 3 respectively. Detection limits 0.01 0.1 Conclusions: Sample Enzytec Citric Acid implemented adjusted, from Trinity Oxalate kit. simultaneous determination resulted in fast cost-effective, high throughput methods.
